Pan-kanser analiziyle desteklenen ve deneysel do\u011frulamalarla g\u00fc\u00e7lendirilen bu \u00e7al\u0131\u015fma, CHRDL1\u2019in hem prognostik bir biyobelirte\u00e7 hem de tedavi hedefi olarak potansiyelini g\u00f6zler \u00f6n\u00fcne serdi.<\/p>\n<p>Ara\u015ft\u0131rmac\u0131lar, The Cancer Genome Atlas (TCGA) ve di\u011fer kamuya a\u00e7\u0131k veri tabanlar\u0131ndan faydalanarak, 33 farkl\u0131 kanser t\u00fcr\u00fcndeki CHRDL1 ekspresyonunu kapsaml\u0131 \u015fekilde inceledi. D\u00fc\u015f\u00fck CHRDL1 d\u00fczeyine sahip hastalar daha k\u00f6t\u00fc klinik sonu\u00e7larla kar\u015f\u0131la\u015f\u0131rken, bu bulgu proteinin prognostik de\u011ferlendirilmelerde kullan\u0131lmas\u0131 i\u00e7in umut verdi. Bu \u015fekilde klinik kararlar\u0131n y\u00f6nlendirilmesi ve d\u00fc\u015f\u00fck CHRDL1 ekpresyonuna sahip hastalar\u0131n daha hedeflenmi\u015f tedavi stratejileriyle ele al\u0131nmas\u0131 \u00f6nem kazan\u0131yor.<\/p>\n<p>CHRDL1\u2019in ba\u011f\u0131\u015f\u0131kl\u0131k mikro\u00e7evresi ile kurdu\u011fu ili\u015fki dikkat \u00e7ekici bulundu. Y\u00fcksek CHRDL1 seviyeleri, CD4\u207a ve CD8\u207a T h\u00fccreleri, B h\u00fccreleri, n\u00f6trofiller, makrofajlar ve dendritik h\u00fccreler gibi ba\u011f\u0131\u015f\u0131kl\u0131k etkin h\u00fccrelerin t\u00fcm\u00f6r i\u00e7ine daha fazla yerle\u015fmesine pozitif olarak ba\u011fl\u0131 bulundu. Bu bulgu, CHRDL1\u2019in anti-t\u00fcm\u00f6r ba\u011f\u0131\u015f\u0131kl\u0131k yan\u0131t\u0131n\u0131 destekleyerek t\u00fcm\u00f6rde ba\u011f\u0131\u015f\u0131kl\u0131k g\u00f6zetimini art\u0131rd\u0131\u011f\u0131na i\u015faret ediyor ve b\u00f6ylece tedaviye yan\u0131t \u015fans\u0131n\u0131 y\u00fckseltebilir.<\/p>\n<p>Daha derin analizler, CHRDL1 y\u00fcksek olan t\u00fcm\u00f6rlerin daha iyi ba\u011f\u0131\u015f\u0131kl\u0131k profillerine sahip oldu\u011funu ve t\u00fcm\u00f6r k\u00f6k h\u00fccre \u00f6zelliklerinin ise azald\u0131\u011f\u0131n\u0131 g\u00f6sterdi. T\u00fcm\u00f6r k\u00f6k h\u00fccreli\u011fi, tedavi direnci ve metastaz ile ba\u011flant\u0131l\u0131yken, CHRDL1\u2019in bu \u00f6zellikle ters orant\u0131l\u0131 olmas\u0131, proteinin t\u00fcm\u00f6r heterojenli\u011fi ve agresifli\u011fini s\u0131n\u0131rland\u0131rd\u0131\u011f\u0131n\u0131 ve ba\u011f\u0131\u015f\u0131kl\u0131k kontrol\u00fcn\u00fc kolayla\u015ft\u0131rd\u0131\u011f\u0131n\u0131 kan\u0131tl\u0131yor.<\/p>\n<p>Bioinformatik analizlerin \u00f6tesinde, ara\u015ft\u0131rmac\u0131lar CHRDL1\u2019i A549 ve H1299 akci\u011fer adenokarsinom h\u00fccre hatlar\u0131nda yapay olarak \u00e7o\u011faltarak do\u011frulama \u00e7al\u0131\u015fmalar\u0131 yapt\u0131. CHRDL1\u2019nin fazlal\u0131\u011f\u0131, h\u00fccre proliferasyonu, g\u00f6\u00e7 ve invazyon kapasitesinde anlaml\u0131 azalmalarla sonu\u00e7land\u0131. CCK-8 testleri ve yara iyile\u015fme ile invazyon deneyleri, CHRDL1\u2019in kanser h\u00fccrelerinin agresif davran\u0131\u015f\u0131n\u0131 do\u011frudan bask\u0131lad\u0131\u011f\u0131n\u0131 fizyolojik olarak destekledi.<\/p>\n<p>In vivo olarak ise ba\u011f\u0131\u015f\u0131kl\u0131k sistemi bask\u0131lanm\u0131\u015f \u00e7\u0131plak farelerde ger\u00e7ekle\u015ftirilen subkutan t\u00fcm\u00f6r olu\u015fum testlerinde CHRDL1 a\u015f\u0131r\u0131 eksprese eden h\u00fccrelerden geli\u015fen t\u00fcm\u00f6rler daha yava\u015f b\u00fcy\u00fcyerek biyobelirtecin t\u00fcm\u00f6r olu\u015fumunu engelleme kapasitesi teyit edildi. Bu deneysel sonu\u00e7lar, CHRDL1\u2019in terap\u00f6tik m\u00fcdahalelerle art\u0131r\u0131lmas\u0131 halinde tedavi avantajlar\u0131 sa\u011flayabilece\u011finin somut kan\u0131t\u0131 oldu.<\/p>\n<p>Pan-kanser yakla\u015f\u0131m\u0131, CHRDL1\u2019in sadece akci\u011fer kanseriyle s\u0131n\u0131rl\u0131 olmayan, \u00e7ok y\u00f6nl\u00fc t\u00fcm\u00f6r biyolojisi \u00fczerindeki rol\u00fcn\u00fc ortaya koydu. Hem t\u00fcm\u00f6r h\u00fccre i\u00e7i mekanizmalar hem de ba\u011f\u0131\u015f\u0131kl\u0131k mikro\u00e7evresi \u00fczerindeki d\u00fczenleyici etkileri, proteini immunoterapi kombinasyonlar\u0131nda kullan\u0131labilecek de\u011ferli bir hedef haline getirdi.<\/p>\n<p>Bununla beraber, \u00e7al\u0131\u015fmada CHRDL1\u2019in BMP sinyal yoluyla ve t\u00fcm\u00f6r mikro\u00e7evresiyle nas\u0131l etkile\u015fti\u011fine dair daha derin mekanik a\u00e7\u0131klamalara ihtiya\u00e7 oldu\u011fu vurguland\u0131.
